《One-step Quickcolor Mycoplasma Detection Kit》 is mainly used to detect whether mycoplasma is contained in cell culture medium or other liquid samples. This product is for research use only. Mammalian cell culture, mycoplasma contamination is a global problem. Mycoplasma contamination can almost change all parameters of cells, leading to inaccurate or even completely wrong experimental results (for detailed hazards of mycoplasma contamination to cells, please refer to the company's website: www.yisemed.com). Since 2013, the journal 《Nature》 has officially required that articles involving cell culture must be tested for mycoplasma. It is believed that more and more high-level journals will make the same mycoplasma testing requirements. Culture method is a relatively reliable mycoplasma detection technology, but this method is very time-consuming, requiring several weeks, and is not suitable for rapid detection of mycoplasma contamination in cell culture medium. In addition, solid culture method cannot detect one of the most common mycoplasmas that contaminate cells, namely Mycoplasma hyorhinis (M.Hyorhinis). This is because Mycoplasma hyorhinis cannot form visible colonies on mycoplasma solid culture medium. Mycoplasma hyorhinis accounts for about 20-50% of all mycoplasma contaminations. Some laboratories use fluorescent staining to detect mycoplasma, but this method has low sensitivity and seriously depends on the experience of the experimenter, and the stability of the experiment is extremely poor. In addition, when the method detects positive results, the cells are often severely contaminated. Although culture method and fluorescent staining are mycoplasma detection methods included in the Chinese Pharmacopoeia, they are not suitable as rapid mycoplasma detection methods due to their obvious shortcomings. The company has currently developed four rapid mycoplasma detection kits with different principles, namely: 《One-step Quickcolor Mycoplasma Detection Kit》, 《PCR Mycoplasma Detection Kit》, 《Luminescent Mycoplasma Detection Kit》 and 《Probe Mycoplasma Detection Kit》, each of which has its own advantages and disadvantages (please refer to the company's website for details). 《One-step Quickcolor Mycoplasma Detection Kit》 uses a unique constant temperature gene amplification technology, which has obvious advantages: (1) The entire detection process only takes 1 hour, which greatly shortens the detection time; (2) It has not been found that PCR inhibitors in cell culture medium will inhibit constant temperature gene amplification, so sample pretreatment is generally not required; (3) The sensitivity of constant temperature gene amplification is good, generally higher than 30 cycles of ordinary PCR method; (4) The products of constant temperature gene amplification do not need electrophoresis, and the reaction results can be directly judged by the color change of the indicator; (5) No PCR instrument, electrophoresis tank, gel imager and other instruments are needed, and the entire detection process only requires a water bath. The main disadvantages of 《One-step Quickcolor Mycoplasma Detection Kit》: (1) The mycoplasma recognition rate is relatively low, and it can only recognize about 99% of all mycoplasma species that contaminate cells, and about 1% of mycoplasma may not be recognized, resulting in false negatives; (2) High concentrations of serum and related blood products, divalent ions and EDTA, etc. may interfere with the color development results (Note: Interference can be removed by centrifugal washing), leading to misjudgment of results. After testing, this kit can detect at least the following 13 mycoplasmas: (1) M. hyorhinis, (2) M. fermentans, (3) M. arginini, (4) M. hominis, (5) M. orale, (6) M. salivarium, (7) M. pirum, (8) A. laidlawii, (9) M. agalactiae, (10) M. bovis, (11) M. buccale, (12) M. arthritidis, (13) M. pulmonis (Note: M. is the abbreviation of Mycoplasma; A. is the abbreviation of Acholeplasma). Among them, the first 8 are the most common mycoplasma species that contaminate cells, accounting for about 98% of the mycoplasma that contaminate cells. The above 13 species account for about 99% of the mycoplasma species that contaminate cells (Note: Currently, the literature reports that there are about 19-20 species of mycoplasma that contaminate in vitro cells. Please refer to the company's 《PCR Mycoplasma Detection Kit》 instructions for specific mycoplasma species). Note: This kit cannot detect Ureaplasma Urealyticum mycoplasma (Note: The probability of this mycoplasma contaminating cells is extremely low)! Detection sensitivity of 《One-step Quickcolor Mycoplasma Detection Kit》: After testing, when the DNA addition amount in each reaction tube is 2 μL, the highest detection sensitivity (ie, the lower limit of detection) of 《One-step Quickcolor Mycoplasma Detection Kit》 is about 20-400 copies, that is, 10-200 copies/μL. The detection sensitivity can be further improved by 10-100 times by centrifugal concentration, extraction of mycoplasma genomic DNA and other measures. Since no rapid mycoplasma detection method can guarantee 100% correct detection results, if you want to obtain 100% correct mycoplasma detection results (for example, customers who carry out cell therapy, customers who perform stem cell culture, customers who produce serum, trypsin, culture medium, customers who sell various primary culture cell lines and tumor cell lines, etc.), choose any two of the 《One-step Quickcolor Mycoplasma Detection Kit》, 《PCR Mycoplasma Detection Kit》, 《Luminescent Mycoplasma Detection Kit》 and 《Probe Mycoplasma Detection Kit》 produced by the company for detection, you will get more satisfactory results.
